    Genome sequence of canine herpesvirus

    Canine herpesvirus is a widespread alphaherpesvirus that causes a fatal haemorrhagic disease of neonatal puppies. We have used high-throughput methods to determine the genome sequences of three viral strains (0194, V777 and V1154) isolated in the United Kingdom between 1985 and 2000. The sequences are very closely related to each other. The canine herpesvirus genome is estimated to be 125 kbp in size and consists of a unique long sequence (97.5 kbp) and a unique short sequence (7.7 kbp) that are each flanked by terminal and internal inverted repeats (38 bp and 10.0 kbp, respectively). The overall nucleotide composition is 31.6% G+C, which is the lowest among the completely sequenced alphaherpesviruses. The genome contains 76 open reading frames predicted to encode functional proteins, all of which have counterparts in other alphaherpesviruses. The availability of the sequences will facilitate future research on the diagnosis and treatment of canine herpesvirus-associated disease

    Genomic and biologic comparisons of cyprinid herpesvirus 3 strains

    Cyprinid herpesvirus 3 (CyHV-3) is the archetypal fish alloherpesvirus and the etiologic agent of a lethal disease in common and koi carp. To date, the genome sequences of only four CyHV-3 isolates have been published, but no comparisons of the biologic properties of these strains have been reported. We have sequenced the genomes of a further seven strains from various geographical sources, and have compared their growth in vitro and virulence in vivo. The major findings were: (i) the existence of the two genetic lineages previously described as European and Asian was confirmed, but inconsistencies between the geographic origin and genotype of some strains were revealed; (ii) potential inter-lineage recombination was detected in one strain, which also suggested the existence of a third, as yet unidentified lineage; (iii) analysis of genetic disruptions led to the identification of non-essential genes and their potential role in virulence; (iv) comparison of the in vitro and in vivo properties of strains belonging to the two lineages revealed that inter-lineage polymorphisms do not contribute to the differences in viral fitness observed; and (v) a negative correlation was observed among strains between viral growth in vitro and virulence in vivo. This study illustrates the importance of coupling genomic and biologic comparisons of viral strains in order to enhance understanding of viral evolution and pathogenesis

    Introducir la perspectiva de género en la docencia universitaria significa analizar situaciones de desigualdad que se pueden dar durante el proceso de enseñanza-aprendizaje. Estamos en los inicios de un proceso largo y en el que participan un movimiento amplio formado por profesorado universitario y personal investigador que juegan un papel importante para reducir o eliminar los sesgos de género en el proceso de formación para combatir la discriminación sexual y la desigualdad de género. La Universidad de Alicante ha desarrollado un ‘Plan de Igualdad de Oportunidades entre Mujeres y Hombres’ y ha creado una Red de Investigación en Docencia Universitaria “Universidad, Docencia, Género e Igualdad” en el marco del proyecto de Redes de investigación desde el Vicerrectorado de Estudios, Formación y Calidad para abordar la cuestión de género en el ámbito universitario. Siguiendo esta línea, desde el Departamento de Trabajo Social presentamos los resultados obtenidos en la red: ‘REDGITS – Red de Estudios de la Docencia en Género e Internacionalización en Trabajo Social’, con el fin de analizar y estudiar la introducción de la perspectiva de género en el Grado en Trabajo Social desarrollando indicadores y recomendaciones para ello

    Alcelaphine herpesvirus 1 (AlHV-1) is a gammaherpesvirus that is carried asymptomatically by wildebeest. Upon cross-species transmission to other ruminants, including domestic cattle, AlHV-1 induces malignant catarrhal fever (MCF), which is a fatal lymphoproliferative disease resulting from proliferation and uncontrolled activation of latently infected CD8+ T cells. Two laboratory strains of AlHV-1 are used commonly in research: C500, which is pathogenic, and WC11, which has been attenuated by long-term maintenance in cell culture. The published genome sequence of a WC11 seed stock from a German laboratory revealed the deletion of two major regions. The sequence of a WC11 seed stock used in our laboratory also bears these deletions and, in addition, the duplication of an internal sequence in the terminal region. The larger of the two deletions has resulted in the absence of gene A7 and a large portion of gene A8. These genes are positional orthologs of the Epstein-Barr virus genes encoding envelope glycoproteins gp42 and gp350, respectively, which are involved in viral propagation and switching of cell tropism. To investigate the degree to which the absence of A7 and A8 participates in WC11 attenuation, recombinant viruses lacking these individual functions were generated in C500. Using bovine nasal turbinate and embryonic lung cell lines, increased cell-free viral propagation and impaired syncytia formation were observed in the absence of A7, whereas cell-free viral spread was inhibited in the absence of A8. Therefore, A7 appears to be involved in cell-to-cell viral spread, and A8 in viral cell-free propagation. Finally, infection of rabbits with either mutant did not induce the signs of MCF or the expansion of infected CD8+ T cells. These results demonstrate that A7 and A8 are both essential for regulating viral spread and suggest that AlHV-1 requires both genes to efficiently spread in vivo and reach CD8+ T lymphocytes and induce MCF

    Prototipagem rápida de um modelo complexo para fabricação de moldes de gesso para barbotina cerâmica

    Computer assisted designing (CAD) is well known for several decades and employed for ceramic manufacturing almost since the beginning, but usually employed in the first part of the projectual ideation processes, neither in the prototyping nor in the manufacturing stages. The rapid prototyping machines, also known as 3D printers, have the capacity to produce in a few hours real pieces using plastic materials of high resistance, with great precision and similarity with respect to the original, based on unprecedented digital models produced by means of modeling with specific design software or from the digitalization of existing parts using the so-called 3D scanners. The main objective of the work is to develop the methodology used in the entire process of building a part in ceramics from the interrelationship between traditional techniques and new technologies for the manufacture of prototypes. And to take advantage of the benefits that allow us this new reproduction technology. The experience was based on the generation of a complex piece, in digital format, which served as the model. A regular 15 cm icosahedron presented features complex enough not to advise the production of the model by means of the traditional techniques of ceramics (manual or mechanical). From this digital model, a plaster mold was made in the traditional way in order to slip cast clay based slurries, freely dried in air and fired and glazed in the traditional way. This experience has shown the working hypothesis and opens up the possibility of new lines of work to academic and technological levels that will be explored in the near future. This technology provides a wide range of options to address the formal aspect of a part to be performed for the field of design, architecture, industrial design, the traditional pottery, ceramic art, etc., which allow you to amplify the formal possibilities, save time and therefore costs when drafting the necessary and appropriate matrixes to each requirement.Projeto assistido por computador (CAD) é bem conhecido por várias décadas e empregado na fabricação cerâmica desde o início, mas usualmente empregado na primeira parte dos processos na concepção do projeto, nem na prototipagem, nem nas fases de fabricação. As máquinas de prototipagem rápida, também conhecidas como impressoras 3D têm a capacidade de produzir, em poucas horas, peças utilizando materiais plásticos de alta resistência, com grande precisão e similaridade com relação ao original, com base em modelos digitais inéditos produzidos por meio de modelagem com softwares de projeto específico ou a partir da digitalização das peças existentes, utilizando os chamados scanners 3D. O principal objetivo deste trabalho é desenvolver a metodologia utilizada em todo o processo de construção de uma parte em cerâmica da inter-relação entre as técnicas tradicionais e novas tecnologias para a fabricação de protótipos. E para aproveitar os benefícios que nos permitem esta nova tecnologia de reprodução. A experiência baseou-se na geração de uma peça complexa, em formato digital, que serviu como modelo. Um icosaedro regular de 15 cm, complexo o suficiente para não sugerir a produção do modelo por meio das técnicas tradicionais cerâmicas (manual ou mecânica). A partir deste modelo digital, um molde de gesso feito na maneira tradicional para colagem por barbotina de massas à base de argila, secagem livre ao ar, queimada e esmaltada de maneira tradicional. Esta experiência mostrou a hipótese de trabalho e abre a possibilidade de novas linhas de trabalho a níveis acadêmicos e tecnológicos que serão explorados em um futuro próximo. Esta tecnologia oferece uma grande variedade de opções para lidar com o aspecto formal de uma peça a ser executada no campo do projeto, arquitetura, projeto industrial, a cerâmica tradicional, a arte cerâmica, etc., que permitem ampliar as possibilidades formais, economizar tempo e, portanto, os custos na elaboração das matrizes necessárias e adequadas para cada exigência.Centro de Tecnología de Recursos Minerales y Cerámic

    Biological Control of the Chagas Disease Vector <i>Triatoma infestans</i> with the Entomopathogenic Fungus <i>Beauveria bassiana</i> Combined with an Aggregation Cue: Field, Laboratory and Mathematical Modeling Assessment

    Current Chagas disease vector control strategies, based on chemical insecticide spraying, are growingly threatened by the emergence of pyrethroid-resistant Triatoma infestans populations in the Gran Chaco region of South America. We have already shown that the entomopathogenic fungus Beauveria bassiana has the ability to breach the insect cuticle and is effective both against pyrethroid-susceptible and pyrethroid-resistant T. infestans, in laboratory as well as field assays. It is also known that T. infestans cuticle lipids play a major role as contact aggregation pheromones. We estimated the effectiveness of pheromone-based infection boxes containing B. bassiana spores to kill indoor bugs, and its effect on the vector population dynamics. Laboratory assays were performed to estimate the effect of fungal infection on female reproductive parameters. The effect of insect exuviae as an aggregation signal in the performance of the infection boxes was estimated both in the laboratory and in the field. We developed a stage-specific matrix model of T. infestans to describe the fungal infection effects on insect population dynamics, and to analyze the performance of the biopesticide device in vector biological control. The pheromone-containing infective box is a promising new tool against indoor populations of this Chagas disease vector, with the number of boxes per house being the main driver of the reduction of the total domestic bug population. This ecologically safe approach is the first proven alternative to chemical insecticides in the control of T. infestans.     GRACy: a tool for analysing human cytomegalovirus sequence data

    Modern DNA sequencing has instituted a new era in human cytomegalovirus (HCMV) genomics. A key development has been the ability to determine the genome sequences of HCMV strains directly from clinical material. This involves the application of complex and often non-standardized bioinformatics approaches to analysing data of variable quality in a process that requires substantial manual intervention. To relieve this bottleneck, we have developed GRACy (Genome Reconstruction and Annotation of Cytomegalovirus), an easy-to-use tookit for analysing HCMV sequence data. GRACy automates and integrates modules for read filtering, genotyping, genome assembly, genome annotation, variant analysis and data submission. These modules were tested extensively on simulated and experimental data and outperformed generic approaches. GRACy is written in Python and is embedded in a graphical user interface with all required dependencies installed by a single command. It runs on the Linux operating system, and is designed to allow the future implementation of a cross-platform version.
